Kyuubei is the sole heir to the Yagyuu family, being the daughter of Yagyuu Koshinori and granddaughter of Yagyuu Binbokusai. Raised as a male according to the samurai code, she is a genius swordsman, surpassing average swordsmen and capable of holding her own against veterans like Hijikata Toushirou and Sakata Gintoki.
During her introduction, she attempted to take Shimura Tae as her fiancée, but was stopped by Shimura Shinpachi and others. Despite this, she still harbors feelings for Tae, maintaining a friendly relationship. Kyuubei often follows Tae around and mimics her activities. She also protects Tae from the stalker Kondou Isao.
