Yukishiro Tomoe was the eldest daughter of a samurai, a low-ranking bureaucrat serving the Tokugawa Shogunate in Edo. Her only sibling was her younger brother Enishi, who saw her as a surrogate parent following their mother's death shortly after his birth. As mandated by the Shogunate, she practiced Buddhism and maintained diaries to document her inner thoughts and emotions, which she found challenging to express openly. This reticence left Kiyosato unaware of the depth of her love for him. Believing he needed greater accomplishments to win her affection, he traveled to Kyoto to join the Mimawarigumi but was killed by the Ishin Shishi assassin, Hitokiri Battōsai (Himura Kenshin), who received a single slash scar on his face during the confrontation. Tomoe met Kenshin shortly after Kiyosato's death and went undercover for some time, pretending not to know that Kenshin had killed her fiancé. Suspicion grew, leading them to leave Kyoto and marry, hiding while Tomoe planned Kenshin's death. When the moment arrived for her to reveal her secrets and kill Kenshin, she realized she had fallen in love with him. To save his life, she sacrificed her own, leaving a second scar across his cheek so he would never forget her love.
